原理
銅綠假單胞菌(又名綠膿桿菌)是革蘭氏陰性桿狀需氧菌。正如其他假單胞菌一樣,銅綠假單胞菌分泌幾種的色素,包括綠膿素(藍綠色)、熒光素(黃綠色,發(fā)熒光)、紅膿素(棕紅色)。King等人開(kāi)發(fā)了假單胞菌瓊脂P(又稱(chēng)King A培養基),用于提高綠膿素和紅膿素的產(chǎn)出,以及假單胞菌瓊脂F(又稱(chēng)King B 培養基)用于提升熒光素的產(chǎn)出【1】。
該顯色培養基用植物源蛋白胨取代動(dòng)物蛋白胨,并基于King 等人的配方而成(熒光混合劑除外)。它是選擇性培養基,用于從膿液、痰和引流液等樣本中分離銅綠假單胞菌。
培養基中的溴棕*銨抑制其他菌群生長(cháng)。它作為季銨類(lèi)化合物和陽(yáng)離子表面活性劑,使銅綠假單胞菌之外的細菌菌體釋放出氮和磷。銅綠假單胞菌裂解熒光底物,使UV下發(fā)射熒光。

配制
46.75 g溶于1000 ml蒸餾水(其中含10ml甘油)。加熱至沸騰使全部溶解,高壓滅菌?;靹蚝蛢A注平板。
培養反應
35-37°C培養18-24小時(shí)后,觀(guān)察如下:
